That's a nice area to visit. Last time I was up was last May, before the crowds arrived, and stayed at Deep Creek (McHenry) Maryland as my base. I've stayed at Wisp Resort and also have rented a lake front house. There are campgrounds in the area too. All good. Haven't done the trip on the bike -- yet.
My microbrew and waterfall itinerary:
A couple miles west of Deep Creek on Sang Run Rd is Mountain State Brewery for lunch.
Mountain State Brewing Co
Just south of the lake, off US-219, is Swallow Falls State park for a short hike and a couple waterfalls.
Further down US-219, through Oakland, past the smallest church in America (worth a five minute stop,) it's up the mountain and past the windmills into West Virginia. Just over the summit, there is a small park at the end of a gravel road on your left, where you'll find a spring which is the head water for the Potomac River. Continue on US-219, then WV-32 to get to Blackwater Falls park. You can can hike to the falls or just take a gander from a perch near one of the park roads.
For dinner, stop in at Blackwater Brewing Company on WV-32, opposite the park entrance road, if memory serves.

To get to Falling Water house, head north on US-219 from Deep Creek, then onto the rolling hills of US-40 west, then right onto PA-381 (at a Pizza Hut?) and through the adventure sports town, Ohiopyle, with its falls. Carry onto Falling Waters further down PA-381. Might want to make a reservation online for the house tour if going on a weekend.
